Mechanisms of Smc5/6-based genome management

 

摘要:

Structural Maintenance of Chromosomes (SMC) complexes are critical for genome managements across species. Among the three eukaryotic SMCs, namely cohesin, condensin, and the Smc5/6 complex (Smc5/6), Smc5/6 has a unique set of roles in regulating genome replication and repair, DNA damage response, and viral DNA restriction. Smc5/6 perturbations can lead to multi-system syndromes in humans and pleiotropies in model organisms. Emerging data suggest that Smc5/6’s core biochemical activities in DNA interactions are pertinent to its diverse roles. We and others found that while Smc5/6 resembles other SMCs in interacting with and sliding along dsDNA, it shows a unique preference for binding to DNA sites where dsDNA and ssDNA are adjacent to each other (junction DNA). These junction sites are frequently formed during DNA replication and recombinational repair, the completion of which is known to require Smc5/6. As such, Smc5/6’s association with ss-dsDNA junctions can provide a unified mechanism to support its multiple genome functions. I will report our progresses in using single molecule approaches to gain insights into how Smc5/6 targets to junction sites, how its multiple subunits contribute to junction binding, and how its junction binding is influenced by other key genome maintenance complexes.
